Notice of intent to negoiate a sole source contract with Oregon State University for NOAA, National Marine Fisheries Service located in Seattle, Washington for Columbia River ocean area mixed-stock analysis the current proposal seeks to collect and genotype
DNA samples, and provide mixed stock analysis (i.e.,
GSI) of fish samples collected between Cape Falcon, Oregon and Ledbetter Point, Washington from
commercial troll and sports fisheries using the CTC microsatellite baseline for genetic assignment. ,
Genetic data is a unique tool that can provide estimates of stock proportions of both wild and hatchery
fish samples from a mixed stock fishery. Samples will be genotyped and arialyzed using GAPS
standardized microsatellite baseline for estimation of mixture proportions . Scale analysis and length
estimates will be used to provide auxiliary age information and will be available for cohort reconstruction.
Each genetics laboratory will be responsible for sending a database with genotypes and associated
sample information to Oregon State University (OSU) for compilation into a final database and genetic
mixture analysis. OSU will send the compiled database to Oregon Department of Fish and Wildlife
(ODFW) for use in ageing analysis. OSU will provide ODFW with regional and individual population
assignments.
Objectives
â The performance period of the contract will be from July 1, 2008 through June 30, 2009
â A report will be provided at the conclusion of the performance period
â Collect and analyze scales to provide auxiliary age estimates
â Compare stock mixtures proportions obtained from analysis of CWT recoveries to proportions
calculated from a GSI analysis on the total collection of genetic samples obtained from each
catch
â Compare stock proportions of legal to sub-legal fish encountered during onboard marine
recreational fisheries sampling in Washington
â For recreational fisheries, compare stock proportions of legal/sub-legal fish sampled by the
onboard observer program 9Washington side) to legal sized fish sampled dockside in Oregon
and Washington.

The Pacific Salmon Commission (PSC) concluded a new Agreement in June of 1999 that established a
new abundance-based management approach to managing the Chinook fisheries subject to the Pacific
Salmon Treaty. The Abundance-Based Management Plan included assignments of new data collection
and analysis tasks to the Pacific Salmon Commission 's Chinook Technical Committee (CTC). The U.S.
Commissioners recommended to Congress that the funding required to carry out these tasks be provided
to state and tribal fishery managers . Congress appropriated $ 1.7 million dollars for this purpose to the
National Marine Fisheries Service (NMFS) in FY 2008.
The U. S. Commissioners invited fishery management agencies , and tribal management groups to submit
proposals to review for funding that would provide new information required for Chinook salmon
abundance-based management. They also directed the USCTC to intensively review these proposals to
determine if they met the program needs and were technically sound.
Based on the USCTC review and recommendation , the Commissioners recommended a series of
proposals for funding , including a proposal from Oregon State University (OSU) to document the mixed
stock analysis of troll and marine sport Chinook fisheries from Cape Falcon North to Ledbetter Point using
GAPS standardized microsatellite genetic markers . This contract addresses item 5b of the December 5,
2006 request for proposals by the CTC; specifically the collection and analysis of mixed stock samples.
This information will improve escapement estimates to CTC standards and improve the data used in
analytical methods that support Chinook salmon abundance based management under the Pacific
Salmon Treaty.
The development of DNA stock separation techniques for Chinook salmon and the establishment of a
coast-wide Chinook Salmon DNA sample baseline have been undertaken by a consortium of laboratories
working in close coordination. It is necessary that the participation of these laboratories is maintained as
this work proceeds. Oregon State University has one of the eight labs that have been involved and Dr.
Michael Banks is a leading expert in this developing field. A sole source contract is justified due to their
special expertise and participation in the development and application of new genetic stock identification
techniques for Chinook salmon.
Based on the U.S. Commissioner's decisions and recommendation , OSU should be sole source
contractor for this project.
